Mechanism of Action
Biotin binds in streptavidin β-barrel central cavity through extensive hydrogen bonding network (8 H-bonds) and hydrophobic contacts. Binding involves conformational closure of flexible loop (L3/4) creating tight fit. Extremely slow dissociation rate (t₁/₂ ~200 days).
