His-tag → Ni-NTA (Affinity Purification) is a bioactive peptide with well-characterized properties and therapeutic applications.
Mechanism of Action
Imidazole nitrogen atoms of histidine residues coordinate with Ni²⁺ ions chelated by NTA (nitrilotriacetic acid) on agarose/agarose beads. Typically 2-3 His residues coordinate each Ni²⁺ in octahedral geometry. Multiple His residues increase avidity. Elution with imidazole (250-500 mM) competes for Ni²⁺ coordination.
