Overview
Insulin glargine is formulated in a clear acidic solution (pH 4.0) using zinc chloride and meta-cresol. Upon subcutaneous injection, the physiological pH causes glargine to precipitate into amorphous microparticles.
Formulation Components
- Active: Insulin glargine (3.5 mg/mL or 10 mg/mL)
- Zinc chloride: Stabilizes hexamers
- Meta-cresol: Antimicrobial preservative
- Phenol: Antimicrobial preservative
Precipitation Mechanism
- Clear acidic solution (pH 4.0) injected subcutaneously
- Neutralization by tissue buffer (pH 7.4)
- Glargine precipitates into amorphous microparticles
- Slow dissolution provides sustained release
- Duration: ~24 hours (U100) or >30 hours (U300)
