Overview

Insulin glargine is formulated in a clear acidic solution (pH 4.0) using zinc chloride and meta-cresol. Upon subcutaneous injection, the physiological pH causes glargine to precipitate into amorphous microparticles.

Formulation Components

  • Active: Insulin glargine (3.5 mg/mL or 10 mg/mL)
  • Zinc chloride: Stabilizes hexamers
  • Meta-cresol: Antimicrobial preservative
  • Phenol: Antimicrobial preservative

Precipitation Mechanism

  1. Clear acidic solution (pH 4.0) injected subcutaneously
  2. Neutralization by tissue buffer (pH 7.4)
  3. Glargine precipitates into amorphous microparticles
  4. Slow dissolution provides sustained release
  5. Duration: ~24 hours (U100) or >30 hours (U300)
