isopeptide cross-linking stability modification
Overview
Isopeptide bonds are non-canonical amide bonds between amino acid side chains, not involving the main chain.
Types
- Lys-Asn/Gln: Found in bacterial surface proteins
- Ubiquitin K48/K63: Protein degradation signals
- Transglutaminase-catalyzed: Lys-Gln cross-links
Biological Significance
Isopeptide bonds in bacterial pili provide extreme mechanical stability. Engineered isopeptide bonds create stable protein fusions (SpyTag/SpyCatcher).
