Overview

Isopeptide bonds are non-canonical amide bonds between amino acid side chains, not involving the main chain.

Types

  • Lys-Asn/Gln: Found in bacterial surface proteins
  • Ubiquitin K48/K63: Protein degradation signals
  • Transglutaminase-catalyzed: Lys-Gln cross-links

Biological Significance

Isopeptide bonds in bacterial pili provide extreme mechanical stability. Engineered isopeptide bonds create stable protein fusions (SpyTag/SpyCatcher).
