Overview

N-glycosylation is a co-translational modification where oligosaccharides attach to asparagine residues in the Asn-X-Ser/Thr sequon.

Mechanism

The process begins in the endoplasmic reticulum with transfer of a preassembled oligosaccharide (Glc3Man9GlcNAc2) to nascent polypeptides by oligosaccharyltransferase.

Types

  • High-mannose: Minimal processing
  • Hybrid: Mixed processing
  • Complex: Extensive Golgi processing

Biological Significance

N-glycosylation affects protein folding, stability, cell-cell recognition, and immune function. IgG Fc glycosylation modulates antibody effector functions.
