Overview
Peptide antibiotics are antimicrobial peptides (AMPs) that kill bacteria through membrane disruption and intracellular targeting.
Classes
- Defensins: Cysteine-rich AMPs in neutrophils and epithelia
- Cathelicidins: LL-37 is the human cathelicidin
- Magainins: Frog-derived AMPs
- Lantibiotics: Bacterially produced modified peptides (nisin)
Mechanism
AMPs typically disrupt bacterial membranes through electrostatic attraction to anionic lipids, forming pores or carpeting the membrane surface.
Clinical Significance
Daptomycin and polymyxins are clinically used AMPs. New AMPs are in development against multidrug-resistant organisms.
