TCR recognizes peptide-MHC complex (pMHC) through complementarity-determining regions (CDRs). CDR1/CDR2 contact MHC helices; CDR3 (most variable) contacts peptide and MHC floor. Binding triggers conformational change in TCR, initiating ITAM phosphorylation via CD3 complex → ZAP-70 → downstream signaling.
