PrPC (predominantly α-helical) converts to PrPSc (β-sheet-rich) through template-directed misfolding. PrPSc acts as seed recruiting PrPC, causing exponential propagation. PrPSc is protease-resistant, forms amyloid plaques. Infectious protein (prion) transmits disease without nucleic acid.
