Mechanism of Action
Ubiquitin conjugated to substrate lysines via E1-E2-E3 enzymatic cascade (isopeptide bond). K48-linked poly-ubiquitin chain (≥4 Ub) is canonical degradation signal. 26S proteasome recognizes poly-Ub via Rpn10/Rpn13 receptors, deubiquitinases recycle Ub, and 20S core particle degrades substrate to peptides.
